Mudumalai National Park is home to several endangered and vulnerable species. It is one of India's few Elephant Reserves and an important elephant corridor for South India. The park is an important habitat for several endangered species like the Indian Vulture, Leopard, Asian Elephant, Golden jackal, Sloth Bear, and many more.

Apart from this there are at least 266 species of birds in the sanctuary, including critically endangered Indian white-rumped vulture and long-billed vulture. Many threatened and regional endemic birds can also be sighted here like the Malabar Trogon, Black-and-orange flycatcher to name a few.
